Born in Kolkata, Madhu Bose was the grandson of historian Romesh Chunder Dutta. He received his early education in Santiniketan and later studied in Vidyasagar College where he met Sisir Bhaduri. Subsequently, he also acted in a Sisir Bhaduri play. In 1923, he joined Madan Theatres as an actor. He also assisted JJ Madan during the making of Turki Hoor (1924). Following a meeting with Himanshu Rai, he started assisting him. Prem Sanyas was one of the major Himanshu films where he worked as an assistant. Here, Madhu was also cast in a small role. Did You Know?

Madhu mentions another documentary called The Musical Instruments of India that he was working on simultaneously. It featured Ustad Vilayat Khan, Ustad Bismillah Khan, Pannalal Ghosh and Venkatagiriyappa. Madhu started editing the film too. However, it could not be completed.
